The aberrant expression and localization of these proteins fail to control DNA repair and apoptosis. sClusterin is the Ku70 partner that sterically inhibits Bax-dependent cell death after damage in some pathologic conditions.

Together, Ku70 and Ku80 make up the Ku heterodimer, which binds to DNA double-strand break ends and is required for the non-homologous end joining (NHEJ) pathway of DNA repair. It is also required for V (D)J recombination, which utilizes the NHEJ pathway to promote antigen diversity in the mammalian immune system . Ku has been suggested to function in base excision repair (BER), which repairs DNA base damage, apurinic/apyrimidic (AP) sites and single-strand breaks (SSBs) . Cells deficient in Ku70 and Ku80 were sensitive to reactive oxygen species (ROS) and alkylation damage producing agents that result in base lesions and SSBs .
